RESUMO

This paper aimed to study the effect of Dalbergia cochinchinensis heartwood on plasma endogenous metabolites in rats with ligation of the left anterior descending coronary artery, and to analyze the mechanism of D. cochinchinensis heartwood in improving acute myocardial ischemic injury. The stability and consistency of the components in the D. cochinchinensis heartwood were verified by the establishment of fingerprint, and 30 male SD rats were randomly divided into a sham group, a model group, and a D. cochinchinensis heartwood(6 g·kg~(-1)) group, with 10 rats in each group. The sham group only opened the chest without ligation, while the other groups established the model of ligation. Ten days after administration, the hearts were taken for hematoxylin-eosin(HE) staining, and the content of heart injury indexes in the plasma creatine kinase isoenzyme(CK-MB) and lactate dehydrogenase(LDH), energy metabolism-related index glucose(Glu) content, and vascular endothelial function index nitric oxide(NO) was determined. The endogenous metabolites were detected by ultra-high-performance liquid chromatography-time-of-flight-mass spectrometry(UPLC-Q-TOF-MS). The results showed that the D. cochinchinensis heartwood reduced the content of CK-MB and LDH in the plasma of rats to relieve myocardial injury, reduced the content of Glu in the plasma, improved myocardial energy metabolism, increased the content of NO, cured the vascular endothelial injury, and promoted vasodilation. D. cochinchinensis heartwood improved the increase of intercellular space, myocardial inflammatory cell infiltration, and myofilament rupture caused by ligation of the left anterior descending coronary artery. The metabolomic study showed that the content of 26 metabolites in the plasma of rats in the model group increased significantly, while the content of 27 metabolites decreased significantly. Twenty metabolites were significantly adjusted after the administration of D. cochinchinensis heartwood. D. cochinchinensis heartwood can significantly adjust the metabolic abnormality in rats with ligation of the left anterior descending coronary artery, and its mechanism may be related to the regulation of cardiac energy metabolism, NO production, and inflammation. The results provide a corresponding basis for further explaining the effect of D. cochinchinensis on the acute myocardial injury.

Leaves are plant structures that express important traits of the environment where they live. Leaf description has allowed identification of plant species as well as investigation of abiotic factors effects on their development, such as gases, light, temperature, and herbivory. This study described populations of Dalbergia ecastaphyllum through leaf geometric morphometrics in Brazil. We evaluated 200 leaves from four populations. The principal component analysis (PCA) showed that the first four principal components were responsible for 97.81% of variation. The non-parametric multivariate analysis of variance (NPMANOVA) indicated significant difference between samples (p = 0.0001). The Mentel test showed no correlation between geographical distances and shape. The canonical variate analysis (CVA) indicated that the first two variables were responsible for 96.77 % of total variation, while the cross-validation test showed an average of 83.33%. D. ecastaphyllum leaves are elliptical and ovate.


As folhas são estruturas presentes nas plantas que expressam importantes características de acordo com o ambiente no qual estão inseridas. Ao longo dos anos, a sua caracterização tem permitido identificar espécies vegetais e correlacionar o efeito de fatores abióticos como gases, luz, temperatura e herbivoria sob o seu desenvolvimento. O presente estudo teve como descrever populações de Dalbergia ecastaphyllum do Brasil utilizando a morfometria geométrica foliar. Foram avaliadas 200 folhas de quatro populações desta espécie. Análise de componentes principais (ACP) mostrou que os quatro primeiros componentes principais explicaram 97,81% da variação. A Análise de variância multivariada com teste não-paramétrico (NPMANOVA) indicou não haver diferença entres as amostras (p= 0,0001). Os resultados do teste de Mentel mostraram que não houve correlação das distâncias geográficas com a forma. Na análise de variação canônica, as duas primeiras variáveis responderam por 96,77 % da variação total, enquanto uma média de 83,33% foi encontrada pelo teste de validação cruzada. As folhas de D. ecastaphullym são elípticas e ovadas.

For the first time, the anti-hemolytic activity and the enzyme inhibitory activities of Dalbergia ecastaphyllum leaves extracts were tested against α-amylase, α-glucosidase, lipase, acetylcholinesterase, butyrylcholinesterase, tyrosinase and hyaluronidase. The phenolic profile of the obtained extracts was also investigated by high-performance liquid chromatography with photodiode array detection (HPLC-PAD). The extracts showed inhibitory activity against all enzymes evaluated, with the highest inhibitory activity reported for the enzyme hyaluronidase (28.28 ± 2.43 to 72.19 ± 1.40 μg mL-1). The obtained extracts also demonstrate anti-hemolytic activity (52.22 ± 1.62 to 71.17 ± 1.82%). Among the phenolic compounds identified, protocatechuic, vanillic and β-resorcylic acids were the most abundant (1.13 ± 0.06 to 2.53 ± 0.06, 0.90 ± 0.06 to 2.19 ± 0.06 and 1.03 ± 1.62 to 22.11 ± 1.62 mg L-1, respectively). In the statistical analysis, a significant correlation was found between the flavonoids content and all enzymes inhibitory activities. The present study showed that D. ecastaphyllum leaves extracts may have the potential to be used in the therapeutic treatment of several diseases such as Alzheimer, Parkinson, type 2 diabetes mellitus, hyperglycemia, and pigmentation, as well as those associated with oxidative stress.

Dalbergiae Odoriferae Lignum is derived from heartwood of Dalbergia odorifera,which is national Ⅱ level of rare and endangered protective plants in China. Its resources are scarce and its price is high. In order to find substitutes of D. odorifera,the chemical constituents of 70% ethanol extract of heartwood of D. catifolia were systematically studied by using silica gel,Sephadex LH-20 column chromatography,and semi-preparative HPLC. Sixteen neoflavanoids were isolated and identified as eight dalbergiphenols( 1-8),three dalbergiones( 9-11),two dalbergins( 12,13),two benzophenones( 14,15) and one other type neoflavanoids( 16) based on spectroscopic data analyses and/or comparing the spectroscopic data with those in literature. Among them,compounds 3,7 and 11 were isolated from the genus Dalbergia for the first time,and compounds 2,4-6,8,14 and 15 were isolated from the D. latifolia for the first time. Ten neoflavonoids were both discovered from D. latifolia and D. odorifera.

RESUMO

The study is aimed to distinguish morphological characteristics of Dalbergiae Lignum collected from crude drug's markets and establish a identification methods and the quality standard for Dalbergiae Lignum. The macroscopic and microscopic features of Dalbergiae Lignum from crude drug's market were observed, analyzed and compared according to Hongmu specification issued by the People's Republic of China in 2000, and by the characteristics recorded in domestic monograph of Mucai Shibie (wood identification). The redwood of Dalbergiae Lignum cut into small pieces as medicinal material are dry heart wood of mahogany (trees from Dalbergia sp.), which characteristics of the small pieces as crude drug are different. There are differences in macroscopic and microscopic features about texture of wood and color, odor, taste, transverse section, radial section, tangential section. The results can provide basis for identification, application and improment of the quality standard of Dalbergiae Lignum as medicinal material.

Dalbergiae Odoriferae Lignum as a traditional Chinese medicine (TCM) has been widely used for promoting blood circulation and removing blood stasis. Flavonoid compounds are main chemical constituents of Dalbergiae Odoriferae Lignum, which exert anti-inflammatory property. However, the underlying anti-inflammatory mechanisms of flavonoid compounds are incompletely understood. It has been reported that isoliquiritigenin, liquiritigenin, naringenin and butein possess anti-inflammatory property. The purpose of this study is to illuminate the anti-inflammatory mechanism of flavonoid compounds based on the protein interaction network (PIN) analysis on molecular network level. 130 targets of the main medicinal ingredients of flavonoid compounds were gained though database retrieval. A protein interaction network of flavonoid compounds was constructed with 589 nodes and 216 interactions. By a graph theoretic clustering algorithm Molecular Complex Detection (MCODE), 26 modules were identified and analyzed by Gene ontology (GO) enrichment. Two modules were associated with anti-inflammatory actions. The most interesting finding of this study was that the anti-inflammatory effect of flavonoid compounds may be partly attributable to inhibite FOS, PTGS2 expression, inhibite of IL-1beta release, and block the MAPK pathway and toll-like receptor pathway.

The mature seeds and excised embryos of Dalbergia odorifera were used as materials to study the effect of moisture content on their survival, as well as the effect of rapid freezing and vitrification freezing method on seeds and in vitro embryos cryopreservation. The results showed that the germination rate and vigor decreased from 82.67%, 85% to 18.35%, 25% respectively, when the seed moisture content decreased from 15.04% to 8.14%; and the germination rate decreased from 82.67% to 37.50%, 25.37% respectively by vitrification freezing method and rapid freezing method, when the seed moisture content decreased from 15.04% to 9.37%. Among all the moisture content gradient, 12.35% moisture reached the maximal germination rate, which were 63.58% and 50.45% respectively by vitrification freezing and rapid freezing; and when the embryo moisture content was 26.32%, the germination rate decreased from 95.67% to 58.31% and 33.82% respectively by vitrification freezing and rapid freezing. And when the moisture content was in the range of 14.17% -21.34%, the germination rate was a bit of decrease. The experiment results showed that the optimum conditions of seed cryopreservation were: moisture content 12.35%, vitrification freezing; and the optimum conditions of in vitro embryo cryopreservation were: moisture 15.04%, vitrification freezing. In conclusion, the effects of moisture content on germination rate after cryopreservation in D. odorifera seeds and embryo were significant, and vitrification freezing method is much better than rapid freezing method.

Fourteen compounds were isolated from Dalbergia odoriferae and purified by repeated column chromatography on silica and sephadex LH-20 gel and structurally identified by spectral analysis. These compounds were identified as 4, 9-dimethoxy-3-hydroxypterocarpan (1), medicarpin (2), 2', 4', 5-trihydroxy-7-methoxyisoflavone (3), 2', 3', 7-trihydroxy-4'-methoxyisoflavan (4), formononetin (5), 3, 8-dihydroxy-9-methoxypterocarpan (6), koparin (7), 3-hydroxy-9-methoxypterocarp-6a-ene (8), 2'-hydroxyformononetin (9), stevenin (10), 2', 7-dihydroxy-4', 5'-dimethoxyisoflavone (11), lyoniresinol (12), 2, 4-dihydroxy-5-methoxy-benzophenone (13) and neokhriol A (14). Compounds 1, 3, 4, 6, 8, 12 and 14 were isolated from this plant for the first time. Antibacterial activity assay showed that compound 4 had inhibitory effect on Ralstonia solanacearum.

O Jacarandá da Bahia é uma espécie florestal nativa pertencente a família Leguminosaea Papilionoideae com ocorrência desde o sul da Bahia até o estado de São Paulo. O extrativismo e a pecuária contribuíram para a sua quase extinção e apesar disso, pouco tem sido feito para a multiplicação desta espécie. A micropropagação é uma alternativa para obtenção de grande quantidade de mudas sadias em curto espaço de tempo. Foram utilizados meristemas apicais e gemas axilares de plântulas de Jacarandá da Bahia, cultivadas em condições assépticas nos tratamentos compostos por meios de cultura (MS e WPM), concentrações de BAP (4, 9 e 18 µM) e com a ausência ou presença de agente antioxidante (PVP ou carvão ativado), totalizando 18 tratamentos, cada um com 10 repetições. As avaliações foram realizadas com 4 e 8 semanas de cultivo, considerando a porcentagem de explantes oxidados. Os meios de cultura de antioxidantes testados não diferiram estatisticamente entre si, porém tratamentos com BAP na concentração de 18 µM foram os que apresentaram maior oxidação.


Dalbergia nigra is a native forest species belonging the Papilionoideae-Leguminosaea family occurring from southern Bahia to the state of Sao Paulo. The extraction and livestock contributed to its near extinction and yet little has been done to the multiplication of this species. The micropropagation techniques are an alternative in order to achieve a large amount of seedlings in a short time and under conditions plant without diseases. Used apical and lateral meristems of Dalbergia nigra seedlings, grown in aseptic conditions in treatments consisting of different culture media (MS and WPM), BAP concentrations (4, 9 and 18 µM) and the absence or presence of antioxidant agent (PVP or activated charcoal), all crossed each other, totaling 18 treatments, each with 10 repetitions. Evaluations were performed at 4 and 8 weeks of cultivation, considering the percentage of browning explants. The culture media of antioxidants tested did not differ statistically, but BAP at a concentration of 18 µM were those with the highest oxidation.

OBJECTIVE@#To investigate the antioxidant and anti-inflammatory activity of orally administered methanolic leaf extract of Dalbergia paniculata (D. paniculata) in Carrageenan induced inflammation in rats.@*METHODS@#In vitro antioxidant activity was evaluated for superoxide radical, Hydroxyl radical and DPPH radical scavenging activity. Three doses 200 mg/kg, 400 mg/kg and 800 mg/kg of D. paniculata were tested for anti-inflammatory activity in Carrageenan induced rat paw edema model and paw thickness was measured every one hour up to 6 h.@*RESULTS@#The methanolic leaf extract of D. paniculata produced dose dependent inhibition of Superoxide radical, Hydroxyl radical and DPPH radicals. In Carrageenan induced inflammation model, all three doses produced significant percentage inhibition of rat paw edema and 800 mg/kg dose produced maximum percent inhibition of rat paw edema (47.83%) at 3h compared to control group.@*CONCLUSIONS@#In the present study we found that methanolic leaf extract of D. paniculata showed good in vitro antioxidant activity and in vivo anti-inflammatory activity in rats.

Dalbergia nigra (Vell.) Fr. All., conhecida popularmente como jacarandá -da-bahia, é uma espécie bastante conhecida pela exploração da sua madeira. Foi realizado um experimento no Laboratório de Análises de Sementes (CCA/UFPB) Areia, Estado da Paraíba, com o objetivo de determinar metodologia adequada para a condução de testes de germinação e vigor com sementes de D. nigra. O experimento foi conduzido em delineamento inteiramente ao acaso, em esquema fatorial 2 x 7, sendo duas temperaturas (25 e 20-30°C) e sete substratos (entre areia, entre vermiculita, bioplant®, plantmax ®, entre e sobre papel mata-borrão e papel toalha), com quatro repetições de 25 sementes cada. As características avaliadas foram: germinação (%), primeira contagem de germinação (%), índice de velocidade de germinação (IVG), comprimento e massa seca de plântulas. As melhores combinações para condução de testes de germinação e vigor com sementes de D. nigra foram: a temperatura de 25°C e os substratos entre vermiculita, sobre papel mata-borrão e papel toalha (organizado na forma de rolo); A temperatura de 20-30°C os substratos Bioplant® e o Plantmax®.


Dalbergia nigra (Vell.) Fr. All., known popularly as jacarandá-da-bahia, it is a plentiful species known for the exploration of its wood. The experiment was conducted at the Laboratory of Seed Analysis (CCA/UFPB), in Areia, Paraíba State, with the objective of determining appropriate methodology for testing germination and vigor of D. nigra seeds. The experiment was carried out in an entirely randomized design in a 2 x 7 factorial scheme, with 2 temperatures (25 and 20- 30ºC) and 7 substrates (between sand, between vermiculite, Bioplant®, Plantmax®, between and on paper blotting paper, and towel paper), with four replications with 25 seeds each. The evaluated characteristics were: germination (%), first germination count (%), germination speed index (GSI), length and dry mass of seedlings. The best combinations for conducting germination and vigor tests with seeds of D. nigra were: temperature of 25°C and the substrate between vermiculite, on paper blotting paper and paper towel (roll); temperature of 20-30°C the substrate Bioplant® and Plantmax®.

<p><b>OBJECTIVE</b>To propose an algorithm of the similarity between UV-spectra for controlling the quality of Chinese medical injection.</p><p><b>METHOD</b>The similarity is calculated base on the data of two UV-spectrum curves, thus the sample quality is evaluated according to the average value of the similarity of the UV-spectra between sample with each of standard samples; the proposed algorithm of the similarity is compared with cosine of vectorial angle and relative coefficient; the proposed method for quality evaluation of traditional Chinese injection is discussed with the method according to the chromatographic fingerprint similarity.</p><p><b>RESULT</b>The calculated similarity with the proposed method can be sensitive to show the difference between UV-spectra. This proposed method for quality evaluation was applied for assessing quality of 14 batches of Xiangdan injection and the results well coincident with sample quality.</p><p><b>CONCLUSION</b>The proposed method can be used to evaluate the quality of the Chinese medical injection according to the similarity of two UV-spectrum curves.</p>

Five published DNA extraction protocols were compared for their ability to produce good quality DNA from fresh and herbarium leaves of several species of the genus Dalbergia. The leaves of these species contain high amounts of secondary metabolites, which make it difficult to perform a clean DNA extraction and thereby interfering with subsequent PCR amplification. The protocol that produced the best DNA quality in most of the Dalbergia species analyzed, utilizes polyvinylpyrrolidone to bind the phenolic compounds, a high molar concentration of NaCl to inhibit co-precipitation of polysaccharides and DNA, and LiCl for removing RNA by selective precipitation. The DNA quality of herbarium specimens was worse than that for fresh leaves, due to collecting conditions and preservation of samples. We analyzed 54 herbarium specimens, but the recovered DNA allowed successful PCR amplification in only eight. For the genus Dalbergia, the herbarium is an important source of material for phylogenetic and evolutionary studies; due to the occurrence of the different species in various geographical regions in Brazil, it is difficult to obtain fresh material in nature. Our results demonstrated that for Dalbergia species the methods used for the collection and preservation of herbarium specimens have a mayor influence on DNA quality and in the success of phylogenetic studies of the species

RESUMO

<p><b>OBJECTIVE</b>To develop a method for determination of nerolidol in the volatile oil of Dalbergia odorifera.</p><p><b>METHOD</b>GC method was used. The samples were separated on Agilent HP-5 column (320 microm x 30 m, 0.25 microm) with the mobile phase of highly pure N2. Flow rate was 2 mL x min(-1).</p><p><b>RESULT</b>Linearity of nemlidol was good linearity in the range of 0.059-1.97 mg x mL(-1), and the average recovery was 97.5%, RSD 2.3%.</p><p><b>CONCLUSION</b>This method is simple, accurate, rapid and reproducible.</p>

<p><b>AIM</b>To establish the method of HPLC-fingerprint analysis for the quality control of Dalbergia odorifera and identify its main constituents by HPLC-MS.</p><p><b>METHODS</b>The 37 hatches of samples were analyzed on a Phenomenex Luna C18 column with a gradient of acetonitrile and 0.3% aqueous acetic acid at a flow rate of 1.0 mL x min(-1) and detected at 275 nm. Furthermore, the typical samples were detected by HPLC-DAD-MS under negative ion mode.</p><p><b>RESULTS</b>37 batches of D. odorifera samples were classified into three types based on the results of similarity analysis. According to the comparison of the tR, MS data and UV maximum absorbance (gamma(max)) values with the standards, 10, 7 and 2 phenolic components were identified in three types of D. odorifera extracts, separately.</p><p><b>CONCLUSION</b>The method is repeatable and reliable, and it is capable of effectively controlling the quality of D. odorifera.</p>

RESUMO

<p><b>OBJECTIVE</b>To study the chemical constituents from the leaves of Dalbergia hainanensis.</p><p><b>METHOD</b>Compounds were isolated by chromatographic techniques on silica gel and polyamide column. Their structures were elucidated by chemical and spectroscopic methods.</p><p><b>RESULTS</b>Thirteen compounds were identified as 8-C-glucosylprunetin (1), 8-C-glucosylgenistein (2), 2-hydroxy-5-methoxy biochanin A (3), formononetin (4), 3,5-dimethoxy-4-hydroxybenzaldehyde (5), 1-O-beta-D-glucopyranosyl-(2S, 3S, 4R, 8Z)-2-[(2R)-2-hydroxyl docosylamino]-8-octadecene-1, 3, 4-triol (6), friedelin (7), taxaxerol (8), 3(-hydroxy-glutin-5-ene (9), ursolic acid (10), beta-sitosterol (11), daucosterol (12), lupeol (13).</p><p><b>CONCLUSION</b>All the compounds were isolated from the plant for the first time.</p>
